Sting-T229A mutation abrogates IFN signaling. A shSTING KYSE-30 cells reconstituted with either STING-WT or STING-T229A were stimulated with HSV-1 (MOI = 1) or transfected with 2 μg/mL poly(dA:dT) by Lipofectamine 2000 for 16 h. Levels of Ifnb1, Il6, Tnfa, Isg15, Cxcl10, and Mx1 mRNA in cells were measured with RT-PCR. B and C shSTING KYSE-30 cells reconstituted with either STING-WT or STING-T229A were stimulated with HSV-1 (MOI = 1) or transfected with 2 μg/mL poly(dA:dT) by Lipofectamine 2000 for 16 h. Levels of IFN-β and IL-6 in the supernatants of the cell culture were measured with ELISA. Data are representatives of 3 independent biological replicates.
