Fig. 3

CXCR2-mediated β-arrestin1/2 recruitment. Concentration-dependent BRET signals were measured upon stimulation of HEK293A cells transfected with CXCR2.mNG and β-arrestin1.NLuc (Panel A) or β-arrestin2.NLuc (Panel B), respectively. Results are presented as a percentage of β-arrestin recruitment, with the maximal CXCL8-induced response set at 100%. Data for the curves represents the Mean ± SEM, while the boxplot depicts the range from minimum to maximum, with a central line indicating the median across five independent experiments. Concentration-response curves were fitted to log (agonist) vs. response (three parameter) in GraphPad V9.3.1 (GraphPad Software, San Diego, CA, USA). Statistical analysis was done using one-way ANOVA followed by a Dunnet’s test. Significant differences were marked with *(P < 0.05), **(P < 0.01), ***(P < 0.01) or ****(P < 0.0001), non-significant differences were marked as ns. The dashed lines represent the mean value of CXCL8
