B56γ3 enhances Akt phosphorylation at both Thr308 and Ser473. A, B Lysates of HeLa, C, D HCT116), and E, F SW480 with vector only (pMSCV), stable B56γ3 overexpression (B56γ3HA or 4HAB56γ3) (A, C, E), control shLuc or stable knockdown of B56γ3 expression (shB56γ3) (B, D, F) were harvested at steady state and analyzed by SDS-PAGE and Western blotting with antibodies as indicated. Representative blots of phospho-Akt (p-Akt) at Thr308 and p-Akt at Ser473 from three independent experiments with similar results were shown. The mean relative expression levels of p-Akt at Thr308 (T308) and Ser473 (S473) of three independent experiments with similar results were quantitated and normalized with total Akt by densitometry. The differences were assessed for statistical significance by two-tailed unpaired student’s t test with p value (*(< 0.05),**(< 0.01), ***(< 0.001)). (n = 3)
