SPARC knockdown downregulates SMAD4 expression. A-C KGN cells were transfected with 50 nM control siRNA (si-Ctrl) or SPARC siRNA (si-SPARC) for 48 h. The mRNA levels of SPARC and TGFB1 (A), ALK5 and TβRII (B), SMAD2, SMAD3, and SMAD4 (C) were examined by RT-qPCR. D KGN cells were transfected with 50 nM control siRNA (si-Ctrl) or SPARC siRNA (si-SPARC) for 48 h. The protein levels of SMAD4 were examined by western blot. The results are expressed as the mean ± SEM of at least three independent experiments. Values that are statistically different from one another (p < 0.05) are indicated by different letters
