TRIM31 is required for autophagy. A The mRNA levels of autophagy-related genes ATG5, AT12, and LC3 were measured in sh-NC and TRIM31-deficient GES-1 cells treated with or without Hp supernatant for 12 h. B The formation of autophagosomes was analyzed by transmission electron microscopy. Representative images and the mean number of autophagosomes per cell (10 cells) were shown. C sh-NC and TRIM31-deficient GES-1 cells were transfected with mCherry-EGFP-LC3-expressing adenovirus. Cells were either unstimulated, treated with 100 ng/ml rapamycin (Rapa) alone, or 100 nM bafilomycin A1 (BafA1) in the presence of 100 ng/ml rapamycin for 8 h. The average number of autophagosome yellow dots (mCherry and EGFP colocalization) and autophagolysosome mCherry dots (EGFP was quenched in acidic lysosome) were determined manually (n = 10 cells).
