From: Endoplasmic reticulum stress downregulates PGC-1α in skeletal muscle through ATF4 and an mTOR-mediated reduction of CRTC2

Fig. 6

mTOR/S6K inhibition prevents the reduction of the expression of CRTC2 caused by ER stress. A CRTC2 protein levels in nuclear extracts from LHCN-M2 myotubes incubated with 250 nM torin 1 and/or 5 µg/ml of tunicamycin for 1 h and B in nuclear extracts from the skeletal muscle of mice treated with torin 1 for 24 h and/or tunicamycin for the last 8 h. CE mRNA levels in LHCN-M2 cultured myotubes transfected with control siRNA, CRTC2 siRNA or S6K siRNA and incubated in the presence or absence (control) of 5 µg/ml of tunicamycin for 2 h. Data are presented as the mean ± SEM (n = 3–5 per group). *p < 0.05, **p < 0.01 and ***p < 0.001 versus control. ##p < 0.01 and ##p < 0.01 versus tunicamycin (A, B) or control siRNA (CE), respectively
