ECFCs can reduce the capacity of T cells to produce pro-inflammatory cytokines. CD3⁺CD25⁻ T cells were co-cultured with HAECs, CB-ECFCs and APB-ECFCs in a fixed 1/6 ratio. After 3 days, T cells were collected, activated with PMA/Ionomycin and then treated with Golgi Plug (a protein transport inhibitor). Intracellular cytokine production was determined in both CD4⁺ (left graphs) and CD8⁺ T cells (right graphs) by flow cytometry. Cells were gated on CD4⁺Foxp3⁻ or CD8⁺Foxp3⁻ T cells. For each marker the gating strategy is indicated on the left and below the figure. Each dot represents a measured value (n = 7 for Tcell alone, n = 6 for HAEC group, n = 8 for Tcells + CB-ECFCs group and n = 7 for Tcells + APB-ECFCs group) collected from 2 independent experiments.
