Adipocyte-derived IL-6 and leptin activate the JAK/STAT3 and AKT signaling pathways to promote PLOD2 expression. a MDA-MB-231 (MB-231) cells were monocultured or cocultured with adipocytes. An inhibitor directed against janus kinase (ruxolitinib) or PBS was added to the culture medium. After 3 days, tumor cells were harvested to detect protein expression levels of PLOD2, P-STAT3 and STAT3. b MDA-MB-231 (MB-231) cells were grown on coverslips in inserts. Cells in the lower chamber were monocultured or cocultured with adipocytes and treated with either ruxolitinib or PBS. After 3 days, cell were fixed and stained for P-STAT3.
