From: The Dictyostelium discoideum RACK1 orthologue has roles in growth and development

Figure 6

D. discoideum cells overexpressing DdRACK1 have growth and developmental defects. (A) Levels of DdRACK1 overexpression. Cell lysates from vegetative AX2, AX2/GFP-DdRACK1 and AX2/GFP-DdRACK1mut strains were analyzed by SDS-PAGE and western blot. DdRACK1 and GFP-fusion proteins at 36 and 66 kDa were detected with polyclonal anti-DdRACK1 antibodies. (B) Growth in shaking suspension of D. discoideum strains. 5 x 104cells/ml were used for inoculation. (C) Growth of D. discoideum strains on lawns of K. aerogenes. Images were taken on days 4 and 5. Scale bar, 1 mm. (D) Bar chart showing diameter of plaques formed by D. discoideum strains in (C) measured between days 4 and 7. The bar represents the mean and SD of ten independent experiments (NS, not significant; P > 0.05). (E) Growth of D. discoideum strains on lawns of E. coli B12 and imaged on days 3 and 6. Scale bar, 1 mm. (F) Bar chart showing diameter of plaques formed by D. discoideum strains in (E) measured between days 4 and 9. The bar represents the mean and SD of ten independent experiments (**P < 0.01; *P < 0.05; NS, not significant; P > 0.05).
