ERK1/2 is involved in PGE2-induced MMP-9 expression. a Cells were treated with or without U0126 (1 μM) for 1 h before exposure to 10 μM PGE2 for the indicated times. After treatment, the conditioned media were collected and analyzed by gelatin zymorgraphy. b Cells were stimulated with PGE2 (10 μM) for the indicated times and the cell lysates were analyzed by Western blot using an antiserum reactive with phospho-ERK1/2 (p-ERK1/2) antibody and membranes were stripped and re-probed with total GAPDH as a control. c Cells were pretreated with or without Sc-19220 (Sc, 3 μM), L798-106 (L798, 3 μM), or GW627368 (GW, 1 μM) for 1 h, and then treated with PGE2 (10 μM) for the indicated times. The conditioned media were collected and analyzed by gelatin zymography.
