Translocation of MMP2 and MMP9 the cytoplasm to the nucleus in response to IL-6. a and b, quiescent MDA-MB-231 cells were treated with and without IL-6 for the indicated time periods, fixed, permeabilized, and immunostained for MMP2 and MMP9 using its specific antibodies followed by probing with Alexa Fluor 488-conjugated secondary antibodies. c, quiescent MDA-MB-231 cells were treated with and without IL-6 for 1 h and the cytoplasmic and nuclear extracts were prepared. An equal amount of protein from the cytoplasmic and nuclear extracts of control and the IL-6-treated MDA-MB-231 cells was analyzed by Western blotting for MMP2 and MMP9 levels using its specific antibodies. The blot was reprobed sequentially with anti-β-tubulin and anti-p53 antibodies to show the relative purity of the cytoplasmic and nuclear extracts
