PTP1B decreases PAF-induced C/EBPß phosphorylation in a GSK-3-dependent manner. iMo-DCs were collected on day 7, incubated for 20 min with 10 μM PTP1B inhibitor, 5 μM SB 216763, alone or in combination, or with their vehicles. Cells were then stimulated with 10 nM PAF for indicated times. Reaction was stopped on ice and cells were collected and lysed. Lysates were separated on SDS-PAGE gels and Western blots were performed using Abs recognizing vinculin, pThr235 C/EBPß and C/EBPß. Control and SB216763-treated cells are shown on two different blots. The blots were stripped between re-blotting with indicated antibodies. Control experiment samples are the same as in Fig. 3a. Representative blots are shown for each experimental condition and compilations are presented as mean ± S.E.M indices of normalized ratios of pThr235 C/EBPß levels calculated as described in Methods, of at least 3 experiments. Significance was established with paired two-way ANOVA with Sidak post-test: *p < 0.05, PTP1B **p < 0,01 vs control cells
