Fig. 5

Waixenicin A transiently mimics the impact of a Mg2+-deficient diet on systemic Mg2+ and Ca2+ homeostasis in vivo. a-d: Mg2+ concentrations in urine, feces, serum and bones from mice fed control (0.05% Mg2+) or Mg2+-deficient (0.003% Mg2+) diet. Mice were sacrificed on day 7, 14, 21, and 28 after feeding. Serum, urine, feces and bone were collected and assessed for Mg2+ (*p < 0.05, **p < 0.01, ***p < 0.001 vs. 0.05% Mg2+ diet; n = 3–6). e-f Ca2+ concentrations assessed in urine and feces from mice in (a-d). No changes were found in Ca2+ levels for serum and bone (data not shown). g-j Mg2+ concentrations in urine, feces, serum and bone from mice intraperitoneally injected once with vehicle (PBS) or waixA (25 mg/kg). Mice were sacrificed on day 1, 2, 3, 5, 7 and 14 after injection. Samples were collected for Mg2+ and Ca2+ analysis (*p < 0.05 vs. vehicle. n = 3). k Ca2+ concentrations in feces from same mice as in (g-j) (*p < 0.05; ***p < 0.001 vs. vehicle; n = 3). No statistically significant difference was found for Ca2+ levels in the samples from urine, serum, or bone (data not shown)
