PMT stimulated release of TNF-α or IL-6 is independent of the mTOR pathway. RAW264.7 cells were stimulated with PMT (5 nM) w or w/o pre-incubation with rapamycin (10 ng/ml) for 1 h. As control, cells were untreated, treated with the solvent control DMSO, with rapamycin or LPS (100 ng/ml) as positive control. After 24 h, supernatants were removed and subjected to ELISA for the measurement of (a) IL-6-, (b) TNF-α-, (c) IL12p40- or (d) IL-10 release.
