Figure 1From: Alterations of Gab2 signalling complexes in imatinib and dasatinib treated chronic myeloid leukaemia cellsInhibitor-dependent Gab2 phosphorylation. (A) K562tet cells stably transfected with the inducible Gab2wt expression vector or ‘empty’ control vector [9] were treated with 1 μg/ml dox for 24 h and then exposed to 1 μM imatinib or 0.01 μM and 1 μM dasatinib or to the same volume of DMSO (vehicle control) for 4 h. Total cellular lysates were analysed by western blotting using the indicated antibodies. (B) The murine proB cell line Ba/F3 was transduced with pBabe-Hygro-Bcr-Abl and subsequently with either pWZL-Bsr-Gab2-HA, or pWZL-Bsr empty control vector [9]. Cells were exposed to either 1 μM imatinib or dasatinib or to the same volume of DMSO (vehicle control) for 1 h. Total cellular lysates were analyzed by western blotting using the indicated antibodies.Back to article page
